Sing puts the target to sleep. It will not affect a target that already has a major status ailment. In Stadium, Sing can't affect a target that has a Substitute.
Generation III and on
Sing's effects remain the same, except that now it can't affect targets with Insomnia, Vital Spirit, or Soundproof as their ability.

Other appearances
Jigglypuff uses Sing as its up B attack in the Super Smash Bros. series. The move puts all ground-bound users near Jigglypuff to sleep; it does not affect players in the air. The more damage the opponent has taken, the longer they sleep.
Swablu sings a soothing melody and transparent circles come out of its mouth. Anything that hears the song and is hit with the shockwave instantly falls asleep.
Kricketune puts its scythes together and rubs them back and forth with each other. Then, its body starts to glow white and golden music notes come out of its body at the opponent. Or Kricketune rubs it scythes back and forth creating multicolored notes.
