Sing puts the target to sleep. It will not affect a target that already has a major status ailment. In Stadium, Sing can't affect a target that has a Substitute.
Generation III and on
Sing's effects remain the same, except that now it can't affect targets with Insomnia, Vital Spirit, or Soundproof as their ability.

Other appearances
Jigglypuff uses Sing as its up B attack in the Super Smash Bros. series. The move puts all ground-bound users near Jigglypuff to sleep; it does not affect players in the air. The more damage the opponent has taken, the longer they sleep.
Swablu sings a soothing melody and transparent circles come out of its mouth. Anything that hears the song and is hit with the shockwave instantly falls asleep.
Kricketune puts its scythes together and rubs them against each other. Then, its body starts to glow white and golden music notes come out of its body at the opponent.
Kricketot dances about and it produces a sound of wood wind chimes. Multicolored music notes then come out from in between its antennae and float around.
In Pokémon Yellow, there is a Trainer in the Pewter City Pokémon Center whose Jigglypuff puts the protagonist'sPikachu to sleep after singing. Although the player's Pikachu will not follow them until they are talked to or until the player exits the Pokémon Center, it will not be inflicted with the Sleep status at any stage.
