Antidote was mentioned in An Undersea Place to Call Home!. Lindsey used one off-screen to cure Ash's Pikachu after he had been badly poisoned by a wild Skrelp's Toxic.
Antidotes first appeared in The Sun, the Scare, the Secret Lair!. Professor Kukui handed one to Ash from his medical kit, for him to cure his Rowlet from the poison inflicted by James's Mareanie's Sludge Bomb.
In The One That Didn't Get Away!, Lana used some Antidote-laced food wrapped around her fishing lure to heal a Kyogre from its poisoning.
In the manga

Pokémon Adventures
Red, Green & Blue arc
In The Secret of Kangaskhan, Red used an Antidote to heal a baby Kangaskhan from her poisoning.
Sword & Shield arc
In Toasty!! Battle Against Toxapex, Henry used an Antidote to heal his Thwackey from the poison inflicted on it by Nessa's Toxapex's Baneful Bunker.

Trivia
- In early appearances, the Antidote was depicted as a vaccine inside of a syringe-type device; this was later changed to being a "spray-type medicine" in Pokémon FireRed and LeafGreen.
